SCP2-ab binding was found to be sigmoidal in concentration and was saturable at a molar ratio of 2:1. Molar ratios of 10:1, 20:1 and 60:1 of sterol to SCP2-ab caused significant and dose-dependent inhibition of SCP2-ab binding, suggesting that the sterol binding site in SCP2-ab is hydrophobic in nature and the binding site is greater than 25 A from the protein surface. The 120-kDa SCP2 isoform (SCP2-b) contains a Ser/Thr cluster (SST) located in the N-terminus, which is phosphorylated in response to P13K activation. It has been suggested that the SST cluster modifies SCP2-b function.
