The results from the first approach lead us to the second approach based on the rationale that miR-328 and miR-328 downregulation protect against cardiac structural remodeling and AF and accordingly, that the overexpression of miR-328 should accelerate cardiac remodeling and promote AF. In the second approach, we injected adenovirus vector carrying the premiR-328 sequence into the right atrial free wall of dogs. On the basis of the data from the first approach, we chose to examine a dog model that had a relatively long AF duration for use in the second approach (Figure III in the online-only Data Supplement). We also decided to coinject AMO-328 into the intramyocardial tissues with adenovirus vector carrying premiR-328. A mere intramyocardial injection of vector caused a statistically significant, but small increase of AF duration in comparison with the injection of vector alone. However, coinjection of vector carrying miR-328 with AMO-328 resulted in a significantly shorter AF duration than that produced by vector alone (Figure III in the online-only Data Supplement). Thus, these results indicate that the forced expression of miR-328 is able to recapitulate the phenotypes of experimental AF in two independent approaches.

To determine whether miR-328 participated in the pathogenesis of AF not only in dogs but also in humans, we analyzed the miR-328 expression level in our previously reported cohort of patients with paroxysmal or persistent AF. The results showed that the expression level of miR-328 was significantly increased in patients with AF compared to control subjects with sinus rhythm (Figure 5 A). Furthermore, the expression levels of miR-328 were significantly increased in patients with AF with underlying structural heart disease (AF-SHD) (Figure 5 B). Importantly, the miR-328 level was significantly higher in patients with persistent AF than in patients with paroxysmal AF ( Figure 5 C).
